Bij de verdeling van het vorstendom Nassau tussen de vijf overgebleven zoons van Jan de Oude in 1607 verwierf Ernst Casimir het graafschap Nassau-Dietz en daarmee is hij de stamvader van die tak. Op jonge leeftijd vertrok hij naar Den Haag om dienst te nemen in het Staatse leger; in 1607 werd hij benoemd tot veldmaarschalk. In 1620 volgde hij zijn oudste broer Willem Lodewijk op als stadhouder van Friesland. Na de dood van zijn neef Maurits van Oranje in 1625 werd hij tevens stadhouder van Groningen en Drente. Hij sneuvelde bij Roermond en werd begraven in Leeuwarden.
